นับแต่นั้นมา มหาวิทยาลัยมหาจุฬาลงกรณราชวิทยาลัย ได้ดำเนินการจัดการศึกษาในรูปแบบมหาวิทยาลัยในระดับอุดมศึกษาโดยมุ่งผลิตบัณฑิตที่มีคุณภาพ สร้างความเป็นเลิศทางวิชาการด้านพระพุทธศาสนา ส่งเสริมการค้นคว้าวิจัยให้การบริการวิชาการแก่สังคมและทำนุบำรุงศิลปวัฒนธรรม มีพระภิกษุสามเณรและคฤหัสถ์ทั่วไปสนใจเข้ามาศึกษาเป็นจำนวนมากตามลำดับ ทำให้อาคารสถานที่ไม่เพียงพอต่อการขยายตัวในเชิงบริหาร

ในปีพุทธศักราช ๒๕๓๕ มหาวิทยาลัยมหาจุฬาลงกรณราชวิทยาลัย ได้ขยายอาคารเรียนไปที่วัดศรีสุดารามวรวิหาร แขวงบางขุนนนท์ เขตบางกอกน้อย กรุงเทพมหานคร ในความอุปถัมภ์ของพระราชพิพัฒน์โกศล (โกศล มหาวีโร) เพื่อรองรับจำนวนพระภิกษุสามเณรและคฤหัสถ์ที่เข้ามาศึกษาในมหาวิทยาลัยเพียงพอ อย่างไรก็ตาม อาคารสถานที่เรียนและหน่วยงานก็ยังมีความจำเป็นต้องขยายเพื่อเหมาะสมและเพียงพอ มหาวิทยาลัยมหาจุฬาลงกรณราชวิทยาลัย จึงปรารภที่จะจัดหาสถานที่เพิ่ม

ในปีพุทธศักราช ๒๕๔๒ นายแพทย์รัศมีและคุณหญิงสมปอง วรรณิสสร ได้มีจิตศรัทธาถวายที่ดินจำนวน ๘๔ ไร่ ๑ งาน ๓๗ ตารางวา เพื่อบูชาคุณสมเด็จพระพุฒาจารย์ (อาสภมหาเถระ) ณ ตำบลลำไทร อำเภอวังน้อย จังหวัดพระนครศรีอยุธยา แก่มหาวิทยาลัยมหาจุฬาลงกรณราชวิทยาลัย เพื่อจัดสร้างศูนย์การศึกษา ในการนี้ สมเด็จพระบรมโอรสาธิราชฯ สยามมกุฎราชกุมาร ทรงพระกรุณาโปรดเกล้าฯ ให้พระเทพโสภณ (ประยูร ธมฺมจิตฺโต) อธิการบดี นำผู้บริหารและผู้มีจิตศรัทธาทั้งสองเข้าเฝ้าเพื่อทูลเกล้าถวายโฉนดที่ดินดังกล่าว โดยเสด็จพระราชกุศลเนื่องในวโรกาสมหามงคลทรงเจริญพระชนมพรรษา ๖ รอบ แห่งพระบาทสมเด็จพระเจ้าอยู่หัวภูมิพลอดุลยเดชมหาราช และมหาวิทยาลัยได้รับพระราชทานที่ดินไปดำเนินการตั้งแต่ปีพุทธศักราช ๒๕๔๒ เป็นต้นมา มหาวิทยาลัยจึงเริ่มจัดทำแผนแม่บทเพื่อพัฒนาที่ดินในบริเวณนี้ให้เป็นสถานที่ตั้งอาคารและเพื่อกำหนดทิศทางการพัฒนาด้านวิชาการด้านกายภาพ และด้านเทคโนโลยีให้มีความเหมาะสมกับภารกิจต่อไปโดยใช้งบประมาณก่อสร้างทั้งสิ้น ประมาณ ๑,๒๐๐,๐๐๐,๐๐๐ บาท (หนึ่งพันสองร้อยล้านบาทถ้วน)

          สำหรับงบประมาณนั้นรัฐบาลได้จัดสรรงบประมาณเพื่อสร้างอาคารเรียนรวมเป็นเงินทั้งสิ้น ๓๙๗,๓๒๕,๐๐๐ บาท (สามร้อยเก้าสิบเจ็ดล้านสามแสนสองหมื่นห้าพันบาทถ้วน) นอกเหนือจากนั้นงบประมาณอีก ๘๐๒,๖๗๕,๐๐๐ บาท (แปดร้อยสองล้านหกแสนเจ็ดหมื่นห้าพันบาทถ้วน) มหาวิทยาลัยต้องดำเนินการเองทั้งหมด ซึ่งขณะนี้กำลังรับบริจาค และจะนำเงินส่วนนี้ไปสร้างอาคารหลังต่าง ๆ ดังกล่าวมา และดำเนินการเพื่อหารายได้ในส่วนที่เหลือ เช่น การทอดผ้าป่ามหากุศล จัดสร้างวัตถุมงคล เป็นต้น

           ในอนาคตมหาวิทยาลัยมีแผนการที่จะซื้อที่ดินขยายเพิ่มเติม โดยวางแผนจะสร้างวัด สถานที่จอดรถ เพื่อให้เพียงพอต่อการขยายตัวของมหาวิทยาลัย ในอนาคต
